How the Numbers Work
You have a 10 mg vial of white powder, a vial of bacteriostatic water, and a syringe marked in units rather than milligrams. The calculator bridges those three objects with one number: concentration. Vial milligrams divided by water millilitres gives mg/mL, and your dose divided by that concentration gives the volume to draw.
Think of cordial in a jug. One bottle of cordial in two litres of water is weaker than the same bottle in one litre, so you pour more of the weak mix to get the same amount of cordial. Melanotan 2 works the same way. The dose is always the dose. What changes with the water is how much liquid carries it.
In numbers: a 10 mg vial with 2 mL of water is 5 mg/mL. A 0.5 mg dose is 0.1 mL, which reads as 10 units on a U-100 insulin syringe; 0.25 mg is 5 units and a 0.1 mg test dose is 2 units. With 1 mL of water the vial is 10 mg/mL and every draw halves, which is why the dosage guide prefers 2 mL: a one-unit slip is 0.05 mg instead of 0.1 mg.
Why the Test Dose Exists
The first injection is the one most people remember, because nausea and flushing peak 30 to 90 minutes after it and the intensity tracks the dose. Protocols reported in the community open at 0.25 mg for three evenings, or 0.1 mg for Type I skin, before moving to the full 0.5 mg. The original phase I study at the University of Arizona dosed by body weight, 0.01 to 0.03 mg/kg, and recorded nausea, stretching and yawning as the dominant effects at the higher end (Dorr et al., Life Sci 1996, PMID 8637402).
The calculator pre-fills that test dose and flags any dose above the reported ceiling for your skin type. A 1 mg first injection at 5 mg/mL is 20 units, four times the usual start, and that is the kind of multiple seen in the published overdose reports.
Loading vs Maintenance
Loading builds melanin. Maintenance keeps it. The daily loading dose is 0.5 mg across skin types once the first three days are tolerated, and what the Fitzpatrick type controls is duration. Type I skin carries MC1R variants that respond weakly to melanocortin signalling, so it loads for three to four weeks. Type IV olive skin reaches its colour in seven to ten days. Types V and VI deepen in under a week.
The cumulative loading figure in the result panel adds three ramp days at the test dose to the remaining days at 0.5 mg, using the midpoint of the reported range. For Type II that is 0.75 mg plus 7.5 mg, about 8 mg from a single 10 mg vial. Reconstituted Melanotan 2 holds for 30 to 40 days in the fridge, so one vial covers loading for every skin type.
Maintenance drops to 0.5 mg at a frequency set by skin type, from twice weekly for Type I down to once every two or three weeks for Types V and VI. Without it the tan fades over four to eight weeks as melanocytes turn over. Some reports use a single 1 mg weekly injection instead of two 0.5 mg doses; the calculator treats that as above the reported per-injection amount and says so, because the side effects on injection day scale with the bolus. The Melanotan 1 vs 2 comparison covers why the MC1R-selective analog follows a different schedule.
Timing and UV Exposure, as Reported
Evening injection is the consistent recommendation across community protocols, for a mechanical reason: the nausea window lands while you are asleep. Melanotan 2 has a half-life of roughly 33 minutes, so the tanning signal comes from daily repetition rather than from the peptide lingering.
UV does not change the dose. It changes how fast the colour appears. The dosage guide reports 10 to 15 minutes of sun during loading as enough to accelerate visible tanning, since the peptide drives melanin production while UV triggers its distribution into the visible skin layers. Longer sessions add burn risk, not pigment. The Melanotan II profile covers the receptor mechanism behind this split.
Safety and Legal Status
Melanotan 2 is not approved by any regulator. The FDA, the UK MHRA, the Australian TGA and the European EMA have all issued consumer warnings, and what is sold online is a research chemical with no manufacturing oversight. The 10 mg on the label is unverified, and this page is an educational tool rather than a protocol to follow.
The named risks are specific. Case reports describe melanoma arising during Melanotan 2 use (Hjuler and Lorentzen, Dermatology 2014, PMID 24355990). Causation is unproven, but the peptide stimulates the same cells that melanoma grows from, and it darkens existing moles in a way that makes dangerous change harder to spot. A dermatologist baseline check and monthly ABCDE self-exams are the minimum precaution reported in the dosage guide.
Nausea affects 80 to 90 percent of users at the start, with facial flushing and drowsiness alongside. In men, MC4R activation can cause spontaneous erections; an erection lasting over four hours is priapism and a medical emergency, and it has been reported after overdose (Devlin and Pomerleau, Clin Toxicol 2013, PMID 23537392). Systemic toxicity with rhabdomyolysis has also been described after injection (Nelson et al., Clin Toxicol 2012, PMID 23121206). A personal or family history of melanoma, many atypical moles, cardiovascular disease, pregnancy and breastfeeding appear on every exclusion list in the literature.

Frequently Asked Questions
How many units is 0.5 mg of Melanotan 2?
It depends on the concentration. A 10 mg vial mixed with 2 mL of bacteriostatic water gives 5 mg/mL, so 0.5 mg is 0.1 mL, which reads as 10 units on a U-100 insulin syringe. With 1 mL of water the vial is 10 mg/mL and the same 0.5 mg is 5 units.
What test dose is reported for a first Melanotan 2 injection?
Community protocols and the site's dosage guide describe 0.25 mg for the first 3 days, or 0.1 mg for Fitzpatrick Type I skin. The small first dose exists because nausea and flushing peak at the start and scale with dose. A 0.5 mg first injection is the most common reason people quit in week one.
How does the loading dose differ by Fitzpatrick skin type?
The daily loading dose is 0.5 mg across skin types once the first 3 days are tolerated. What changes is duration: Type I loads for 3 to 4 weeks, Type II for 2 to 3 weeks, Type III for 1 to 2 weeks, Type IV for 7 to 10 days, and Types V and VI for 5 to 7 days.
What is the maintenance dose for Melanotan 2?
Reported maintenance is 0.5 mg per injection, with frequency set by skin type: twice weekly for Type I, once or twice weekly for Type II, once weekly for Type III, every 10 to 14 days for Type IV, and every 2 to 3 weeks for Types V and VI. The tan fades over 4 to 8 weeks without it.
How many doses are in a 10 mg vial of Melanotan 2?
At 0.5 mg per injection a 10 mg vial holds 20 doses. At 0.25 mg it holds 40. Reconstituted Melanotan 2 keeps for roughly 30 to 40 days refrigerated at 2 to 8 degrees C, so a 10 mg vial covers a full loading phase for most skin types with powder to spare for maintenance.
Is Melanotan 2 legal or FDA approved?
No. Melanotan 2 is not approved by the FDA, the UK MHRA, the Australian TGA, or the European EMA, and all four have issued consumer warnings. It is sold as a research chemical with no manufacturing oversight, so vial content and purity are unverified. This calculator is an educational tool, not a prescription.
What are the main risks reported with Melanotan 2?
Nausea in 80 to 90 percent of users, facial flushing, darkening of moles and freckles, and spontaneous erections in men. Published case reports describe melanoma arising during use (PMID 24355990), priapism after overdose (PMID 23537392), and rhabdomyolysis with systemic toxicity (PMID 23121206). A dermatologist mole check before starting is the baseline precaution.
Does UV exposure change the dose?
The dose stays the same. What UV changes is how fast the tan shows. The dosage guide reports that 10 to 15 minutes of sun during loading accelerates visible color, because UV triggers melanin distribution while Melanotan 2 drives melanin production. Sunburn is counterproductive and adds skin cancer risk on top of the peptide's own.
